nOPV2

nOPV2 is a live attenuated oral poliovirus vaccine specifically engineered to address the risk of circulating vaccine-derived poliovirus type 2 (cVDPV2) outbreaks. It is a modified version of the monovalent oral polio vaccine type 2 (mOPV2), developed by introducing genetic changes to enhance phenotypic and genetic stability and reduce the risk of reversion to neurovirulence. The modifications were made using advanced genetic engineering techniques on a Sabin-2 infectious cDNA clone, resulting in strains less likely to mutate and cause paralysis. nOPV2 was first authorized for use under the World Health Organization’s Emergency Use Listing in November 2020 and has since been deployed widely in outbreak settings.
