Overview

Norcocaine is a pharmacologically active minor metabolite of cocaine, primarily formed in the liver through N-demethylation by cytochrome P-450 enzymes and FAD-containing monooxygenases. It retains central nervous system stimulant properties similar to cocaine and contributes to its overall stimulant and toxic effects, including cardiovascular, neurological, and hepatotoxic risks. Norcocaine has also been shown to possess a higher local anesthetic potential than cocaine. While not typically used recreationally, its presence in biological samples serves as a significant biomarker for recent cocaine use and metabolism, and it is often synthesized from cocaine for research purposes.
