norMitoMet is a mitochondria-targeted small molecule primarily utilized as a negative control in preclinical research evaluating the efficacy of mitochondrially targeted metformin (MitoMet). Structurally, it consists of a triphenylphosphonium (TPP+) cation conjugated to a decyl alkyl chain, but it notably lacks the biguanide functional group essential for metformin's pharmacological activity. Developed by researchers at the Medical College of Wisconsin, norMitoMet is used in studies of pancreatic and breast cancers to demonstrate that the potent anti-tumor effects of MitoMet—such as the inhibition of mitochondrial Complex I and the subsequent disruption of cellular bioenergetics—are specifically due to the mitochondrial delivery of the biguanide moiety rather than the TPP-alkyl vector itself. Unlike MitoMet, norMitoMet does not significantly inhibit mitochondrial respiration or suppress tumor growth in animal models at comparable doses.
