Overview

NOSH-1 is a synthetic hybrid molecule derived from aspirin that is designed to simultaneously release nitric oxide (NO) and hydrogen sulfide (H2S). Developed by researchers at the City University of New York Medical School, it represents a new class of anti-inflammatory and anti-neoplastic agents known as NOSH compounds. By incorporating both NO- and H2S-releasing moieties into the aspirin scaffold, NOSH-1 exhibits significantly enhanced potency compared to traditional aspirin, with IC50 values for cancer cell growth inhibition in the low nanomolar range. Its mechanism of action involves the inhibition of cyclooxygenase enzymes (COX-1 and COX-2), leading to reduced prostaglandin E2 (PGE2) levels, alongside the synergistic cytoprotective and signaling effects of NO and H2S. Preclinical studies have demonstrated its efficacy across various cancer cell lines (including colon, breast, and pancreas) and in animal models of inflammation.
