NOSH-2 is an aspirin-based hybrid molecule designed to simultaneously release nitric oxide (NO) and hydrogen sulfide (H2S). Developed by researchers at the City University of New York, it belongs to the NOSH (nitric oxide- and hydrogen sulfide-releasing) class of non-steroidal anti-inflammatory drug (NSAID) derivatives. These compounds are engineered to maintain the anti-inflammatory efficacy of traditional NSAIDs while significantly enhancing their anti-proliferative potency and potentially improving their safety profile. In preclinical studies, NOSH-2 has demonstrated potent growth inhibition across multiple human cancer cell lines, including colon, breast, pancreatic, lung, and prostate cancers, with IC50 values in the nanomolar to low micromolar range. Its mechanism of action involves the inhibition of COX-1 and COX-2 enzymes and the modulation of cellular signaling through the controlled release of NO and H2S.
