NOSH-3 is a synthetic hybrid molecule designed to release both nitric oxide (NO) and hydrogen sulfide (H2S). It is part of a class of compounds known as NOSH-NSAIDs, specifically an aspirin-based derivative. Developed by researchers at the City University of New York, NOSH-3 was engineered to improve the safety and potency of traditional non-steroidal anti-inflammatory drugs (NSAIDs). By incorporating NO- and H2S-releasing moieties into the aspirin scaffold, the compound aims to reduce the gastrointestinal and cardiovascular toxicity associated with NSAIDs while enhancing anti-inflammatory and anti-proliferative activities. In preclinical studies, NOSH-3 has demonstrated significant growth inhibition across various human cancer cell lines, including colon, breast, pancreatic, and lung cancers, often with potencies in the nanomolar range. It also exhibits anti-inflammatory properties by inhibiting cyclooxygenase (COX) enzymes and reducing prostaglandin E2 (PGE2) levels.
