NOSH-4 is a member of the NOSH (nitric oxide- and hydrogen sulfide-releasing) class of hybrid non-steroidal anti-inflammatory drugs (NSAIDs). Developed by researchers at the City University of New York, NOSH-4 is an aspirin-based compound designed to simultaneously release nitric oxide (NO) and hydrogen sulfide (H2S). This dual-release mechanism is intended to enhance anti-inflammatory and anti-proliferative potency while potentially reducing the gastrointestinal and renal toxicity associated with traditional NSAIDs. In preclinical studies, NOSH-4 and its analogs have demonstrated significant growth inhibition across various human cancer cell lines, including colon, breast, pancreatic, and lung cancers, with IC50 values in the nanomolar range. It also exhibits anti-inflammatory activity through the inhibition of cyclooxygenase enzymes (COX-1 and COX-2) and reduction of prostaglandin E2 (PGE2) levels.
