Overview

NOSH-aspirin (NBS-1120) is a novel, orally active synthetic small molecule hybrid of aspirin (acetylsalicylic acid) designed to release both nitric oxide (NO) and hydrogen sulfide (H2S). Developed by the City University of New York and licensed to Avicenna Pharmaceuticals, NOSH-aspirin is designed to offer enhanced anti-inflammatory, analgesic, antipyretic, and antiplatelet properties with a significantly improved gastrointestinal safety profile compared to traditional aspirin. In preclinical studies, NOSH-aspirin has demonstrated potent antineoplastic activity against various cancer types, including colorectal, breast, pancreatic, lung, and prostate cancers, as well as leukemia. Its mechanism of action involves the inhibition of cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2), the induction of reactive oxygen species (ROS), and the downregulation of oncogenic transcription factors such as FOXM1 and NF-κB, leading to cell cycle arrest and apoptosis. Additionally, it has shown neuroprotective effects in models of neurodegenerative diseases like Parkinson's and Alzheimer's disease by reducing neuroinflammation.
