Overview

NOSO-2G is a preclinical, second-generation antibiotic candidate of the odilorhabdin class being developed primarily by Nosopharm in collaboration with Evotec for the treatment of severe nosocomial Gram-negative infections, particularly hospital-acquired pneumonia and ventilator-associated pneumonia caused by carbapenem-resistant Pseudomonas aeruginosa and other multidrug-resistant pathogens.[2][3][7][11] Odilorhabdins are novel ribosome-targeting antibiotics originally derived from Xenorhabdus bacteria; they inhibit bacterial protein synthesis by binding to a unique site on the 30S ribosomal subunit that is distinct from existing ribosome-targeting antibiotics, providing a new mechanism of action against multidrug-resistant Gram-negative bacteria.[1][2][10][12] Within the GNA NOW IMI AMR Accelerator consortium, NOSO-2G was pursued as a lead-optimization programme to select a clinical candidate, but the programme received a no-go decision and was terminated within GNA NOW in mid‑2022 due to unmet criteria and limited medicinal chemistry resources, although further development is being pursued outside that framework.[2][5]
