NOSO-95179 is a synthetic nonapeptide antibiotic of the odilorhabdin class, originally derived from the natural products produced by the entomopathogenic bacterium Xenorhabdus nematophila, and developed by Nosopharm as a lead compound for multidrug-resistant Gram-negative infections.[1][3][8][11] Odilorhabdins bind to the bacterial ribosome at a novel site and inhibit translation with a distinct mode of action from existing ribosome-targeting antibiotics, leading to bactericidal activity against Enterobacteriaceae, including carbapenem-resistant strains such as NDM-1-producing Klebsiella pneumoniae and Escherichia coli.[1][2][5][6][8][10] Structure–activity relationship studies on NOSO-95179 optimized amino acid positions to improve inhibition of bacterial translation and in vitro potency, and the compound demonstrated clear in vitro activity against multidrug-resistant clinical isolates and promising efficacy in mouse models of K. pneumoniae septicemia and lung infection, with low toxicity.[2][5][8][10][12][13]
