notch1 mAb refers to a class of monoclonal antibodies designed to target the Notch1 receptor, a transmembrane protein involved in cell-fate determination that is frequently mutated or overexpressed in various malignancies. These antibodies, such as the one described by Pfizer researchers, typically bind to the negative regulatory region (NRR) of the human Notch1 receptor. This binding prevents the proteolytic cleavage of the receptor by gamma-secretase, thereby blocking the release of the Notch1 intracellular domain (NICD) and inhibiting the activation of downstream target genes like Hes-1 and cMyc. In preclinical models, Notch1 mAbs have demonstrated robust anti-tumor efficacy in T-cell acute lymphoblastic leukemia (T-ALL) and solid tumors such as breast cancer by inducing cell cycle arrest and apoptosis. While several candidates have entered clinical trials, development has often been challenged by gastrointestinal toxicities associated with pan-Notch inhibition.
