notch1 siRNA is a double-stranded RNA molecule designed to silence the expression of the Notch1 gene through the RNA interference (RNAi) pathway. Notch1 is a transmembrane receptor that plays a pivotal role in the Notch signaling pathway, which is frequently dysregulated in various cancers, including glioblastoma multiforme (GBM). In GBM, Notch1 signaling is associated with the maintenance of glioma-initiating cells (GICs), which contribute to tumor recurrence and resistance to standard therapies. By inducing the sequence-specific degradation of Notch1 mRNA, this siRNA reduces the stemness of GICs and enhances the efficacy of co-administered treatments. Research has explored its delivery via specialized vehicles, such as GIC-targeting exosomes, often in combination with other agents like mitoxantrone and interleukin-12, to reshape the immunosuppressive tumor microenvironment and prevent post-surgical recurrence.
