Overview

Notch1 small interfering RNA (siNotch1) is a synthetic, double-stranded RNA molecule designed to silence the expression of the Notch1 gene through the RNA interference (RNAi) pathway. Notch1 is a transmembrane receptor that plays a critical role in cell fate determination, proliferation, and survival; its aberrant activation is frequently associated with tumorigenesis and the maintenance of cancer stem cells (CSCs). By specifically targeting Notch1 mRNA for degradation, this siRNA reduces Notch1 protein levels, thereby inhibiting downstream signaling, inducing apoptosis, and suppressing tumor growth. In experimental settings, it has been investigated as a monotherapy and in combination with oncolytic viruses like H101 to enhance therapeutic efficacy in various cancers, including cervical cancer, lung cancer, and uveal melanoma. The siRNA sequence used in specific studies (e.g., 5'-AAG GUG UCU UCC AGA UCC UGA-3') has demonstrated the ability to inhibit tumor proliferation and promote viral replication of p53-targeted oncolytic agents.
