Overview

NOUS-209 is an investigational, off-the-shelf, neoantigen-targeted cancer vaccine developed by Nouscom. It is designed for the treatment and interception of tumors characterized by mismatch repair deficiency (dMMR) and high microsatellite instability (MSI-H), such as colorectal, gastric, gastro-esophageal junction, and urothelial cancers, as well as for cancer interception in Lynch syndrome carriers. The vaccine utilizes a heterologous prime-boost regimen consisting of a Great Ape Adenovirus vector (GAd20-209-FSP) for priming and a Modified Vaccinia Ankara vector (MVA-209-FSP) for boosting. These viral vectors are engineered to encode 209 shared frameshift peptide (FSP) neoantigens that are commonly expressed in MSI-H/dMMR tumors but absent in healthy tissues. By delivering these shared neoantigens, NOUS-209 trains the host immune system to mount a robust, broad, and durable T-cell response (both CD4+ and CD8+) to recognize and eliminate precancerous and cancerous cells before tumors can fully develop or progress.
