Overview

NOUS-PEV is a personalized, vector-based cancer immunotherapy developed by Nouscom. It is designed for each patient based on the selection and prioritization of mutations unique to that patient's tumor, using next-generation sequencing and proprietary algorithms. The therapy uses a heterologous prime/boost platform composed of two viral vectors: a non-human Great Ape Adenoviral vector (GAd20) for priming and Modified Vaccinia Ankara (MVA) for boosting. Each vector encodes up to 60 patient-specific neoantigens, aiming to induce broad and potent anti-tumor T cell responses by expanding memory CD8+ T cells specific to the tumor's unique mutations. The vaccine is administered in combination with pembrolizumab (an anti-PD1 checkpoint inhibitor) in clinical trials targeting advanced melanoma and non-small cell lung cancer with high PD-L1 expression[1][2][3][4][5][6].
