NOV004 is a peptide drug conjugate engineered to concentrate at the site of bone fracture and deliver a therapeutic anabolic peptide, resulting in rapid and robust bone healing with increased bone density and accelerated repair. It is designed for targeted bone regeneration in rare skeletal diseases, bone injury, and bone cancer, with a lead indication in osteogenesis imperfecta. NOV004 achieves high local concentrations at the fracture site, prolongs therapeutic exposure, improves bone production (up to three-fold compared to non-targeted therapies), and demonstrates enhanced bone strength and density in preclinical models. The drug was originally developed by Novosteo and further advanced within Quince Therapeutics' pipeline after the acquisition of Novosteo by Cortexyme and subsequent rebranding as Quince Therapeutics. IND-enabling studies have been completed, and clinical development for osteogenesis imperfecta is anticipated.
