Drug intelligence / Profile preview
Novel allosteric agonist antibodies targeting integrin α3β1 (alpha-3 beta-1) are being developed by Allosite Therapeutics (formerly 149 Bio) in collaboration with Rush University Medical Center for the treatment of glomerular kidney diseases, including focal segmental glomerulosclerosis (FSGS). Integrin α3β1 is highly expressed on kidney podocytes and is essential for maintaining their attachment to the glomerular basement membrane (GBM). Progressive loss of podocytes due to de-adhesion and injury is a hallmark of FSGS and other glomerular diseases. These antibodies act as allosteric agonists, binding selectively to the α3 head-domain away from the ligand-binding pocket to stabilize the active conformation of the integrin. This activation enhances cell adhesion, strengthens the F-actin cytoskeleton, and protects podocytes from injury-induced detachment and damage.
