Novel HDAC 8 Inhibitor 1 (likely identified as WK2-16) is an experimental small molecule inhibitor of histone deacetylase 8 (HDAC8) currently in preclinical development for the treatment of glioblastoma (GBM). Developed through a collaboration between Sookmyung Women’s University and Yonsei University, the compound was designed to address the limited efficacy of previous HDAC inhibitors in GBM trials. Preclinical studies using human-derived GBM tumorspheres (TS) have demonstrated that the inhibitor reduces cell viability, ATP levels, and the expression of proteins associated with stemness and invasiveness. In vivo studies in mouse models have shown that treatment with this novel HDAC8 inhibitor leads to significant improvements in survival rates compared to controls and standard treatments like SAHA (vorinostat).
