Drug intelligence / Profile preview
Novel oral poliovirus vaccine type 2 (nOPV2) is a next-generation, live-attenuated oral polio vaccine developed to address outbreaks of circulating vaccine-derived poliovirus type 2 (cVDPV2). It is a genetically modified version of the Sabin monovalent oral polio vaccine type 2 (mOPV2), engineered with five specific modifications (including changes in the 5' untranslated region, the capsid protein coding region, the non-structural protein 2C, and the polymerase 3D) to enhance its genetic stability and significantly reduce the risk of reversion to neurovirulence. nOPV2 was developed by a global scientific consortium including the National Institute for Biological Standards and Control (NIBSC), the US Centers for Disease Control and Prevention (CDC), the US Food and Drug Administration (FDA), and the University of California, San Francisco (UCSF), with funding from the Bill & Melinda Gates Foundation. It is manufactured by PT Bio Farma in Indonesia and Biological E in India. nOPV2 received WHO Emergency Use Listing (EUL) in November 2020 and achieved full WHO prequalification in December 2023, serving as a critical tool in global polio eradication efforts.
