Novel oral poliovirus vaccine type 2 candidate 2 (nOPV2-c2) is a live-attenuated vaccine developed to combat circulating vaccine-derived poliovirus type 2 (cVDPV2) outbreaks. Derived from a modified Sabin 2 infectious cDNA clone, it incorporates specific genetic modifications, including S15 domain V changes and CpG40, designed to enhance the stability of its attenuated phenotype. This engineering aims to significantly reduce the vaccine's propensity to revert to neurovirulence, thereby lowering the risk of generating new cVDPV2 strains compared to conventional oral poliovirus vaccines. The vaccine works by inducing a robust immune response against poliovirus type 2. Its development was a collaborative effort involving a consortium led by the Bill & Melinda Gates Foundation, the U.K. National Institute for Biological Standards and Controls, the U.S. Centers for Disease Control and Prevention, PATH, and the University of California San Francisco. While its counterpart, nOPV2 candidate 1, received WHO Emergency Use Listing and Prequalification, nOPV2-c2 was not advanced for further clinical development or Emergency Use Listing due to failing to meet non-inferiority criteria for immunogenicity at lower doses during Phase II trials.
