Novel PKRi is a preclinical-stage small molecule inhibitor of protein kinase R (PKR), also known as EIF2AK2, being developed by Transposon Therapeutics. The program targets PKR pathways that are pathologically activated by the expression of LINE-1 (Long Interspersed Nuclear Element-1) retrotransposons. LINE-1 activation is associated with neuroinflammation and the cellular stress response, which are implicated in the pathogenesis of various cognitive disorders. By inhibiting PKR, Transposon Therapeutics aims to mitigate these downstream effects and provide a therapeutic benefit for disorders of cognition. The asset is currently in the discovery and preclinical development phase.
