Overview
novel small-molecule TL1A antagonist is an orally active, small-molecule allosteric antagonist of tumor necrosis factor-like ligand 1A (TL1A; TNFSF15) being developed by IntelliGen Therapeutics. It is designed for the treatment of immune-mediated inflammatory diseases (IMIDs), such as ulcerative colitis and Crohn's disease. The compound acts via an allosteric mechanism, binding to and stabilizing the TL1A cytokine trimer in an inactive state. This prevents TL1A from interacting with its receptor, death receptor 3 (DR3; TNFRSF25), thereby blocking downstream pro-inflammatory signaling pathways. The program is currently in preclinical development.
