NovoTTF-100L + pemetrexed + cisplatin is a combination therapy consisting of three components: - **NovoTTF-100L**, a medical device that delivers Tumor Treating Fields (TTFields)—low-intensity, alternating electric fields tuned to specific frequencies—to disrupt mitosis of cancer cells. - **Pemetrexed**, a multi-targeted antifolate chemotherapeutic agent that inhibits several folate-dependent enzymes involved in nucleotide synthesis, leading to the inhibition of DNA and RNA synthesis. - **Cisplatin**, a platinum-based chemotherapeutic agent that causes cross-linking of DNA, resulting in apoptosis and inhibition of cell proliferation. This combination is indicated as a first-line treatment for adult patients with unresectable, locally advanced or metastatic malignant pleural mesothelioma. NovoTTF-100L is the first FDA-approved device for mesothelioma in over 15 years and is used in conjunction with pemetrexed and a platinum-based chemotherapy, primarily cisplatin. The mechanism of the combination addresses cancer cell division through electrical field disruption and DNA synthesis inhibition, providing a non-invasive and antimitotic approach[1][5][7].
