Overview

NOX66 + doxorubicin is an investigational combination therapy that merges **NOX66** (a proprietary formulation of idronoxil, delivered as a rectal suppository) with **doxorubicin** (an anthracycline chemotherapy agent). NOX66 acts primarily as a chemosensitizer, aiming to enhance the efficacy of cytotoxic agents such as doxorubicin by inhibiting the tumor cell enzyme ENOX2 (ectonoxotase 2), leading to increased tumor sensitivity to chemotherapy. Doxorubicin intercalates DNA and inhibits topoisomerase II, causing tumor cell death. The combination is being developed for advanced, refractory solid tumors, particularly when standard treatments have failed. The main developer of NOX66 is Noxopharm. Most data currently published explore NOX66 with carboplatin, with rationale and preclinical strategy supporting combinations with other chemotherapeutic agents, including doxorubicin.
