Overview

NP-343 is an orally available small molecule inhibitor of soluble epoxide hydrolase (sEH, also known as EPHX2) being developed by NeuroPn Therapeutics. It belongs to a series of piperidine urea derivatives designed to treat chronic neuropathic pain, neurodegenerative diseases, and potentially oncology. The mechanism of action involves inhibiting the sEH enzyme, which normally degrades protective lipid mediators called epoxyeicosatrienoic acids (EETs) into inactive dihydroxyeicosatrienoic acids (DHETs). By stabilizing endogenous EET levels, NP-343 stimulates GABAergic and opioid pathways, providing non-narcotic, disease-modifying pain relief and neuronal protection. The compound is currently in preclinical development for indications such as diabetic peripheral neuropathy, chemotherapy-induced peripheral neuropathy, and Parkinson's disease (where the related candidate NP-319 is also being explored).
