Overview

**NP-Act D** is a nanoparticle-based formulation of actinomycin D (Act D), an antibiotic-derived antineoplastic agent, designed for controlled, slow release to minimize the severe toxicities associated with standard intravenous Act D. Actinomycin D binds to DNA, inhibiting RNA synthesis by preventing RNA polymerase elongation, particularly effective against transcription in guanine-cytosine-rich regions. Developed by Tiziana Life Sciences, it shows potential for treating **myelodysplastic syndrome (MDS)**, **acute myeloid leukemia (AML)** (especially NPM1-mutated), various solid tumors, and autoimmune/inflammatory diseases like NASH, ulcerative colitis, and rheumatoid arthritis via T-cell modulation; animal studies demonstrate sustained pharmacokinetics over 32 hours and superior tolerability with 0% mortality versus >90% for free Act D.[1][6]
