Overview

NP-TRIMEL is a nanoparticle-based therapeutic cancer vaccine formulation currently in pre-clinical development for the treatment of melanoma. It consists of TRIMEL—a heat-shocked lysate derived from three human melanoma cell lines—encapsulated within poly(lactic-co-glycolic acid) (PLGA) nanoparticles using a double-emulsion method. This formulation was designed to address the stability and storage limitations of earlier tumor lysate vaccines (such as TRIMELVax), which require ultra-low temperatures. NP-TRIMEL functions by delivering tumor-associated antigens to antigen-presenting cells, specifically promoting the generation of TAPCells (dendritic cells) and inducing a robust cytotoxic T lymphocyte response against melanoma cells. The PLGA encapsulation significantly enhances the immunogenicity and stability of the antigens, maintaining functional efficacy for up to 24 weeks at 4 °C.
