NP137 is a first-in-class humanized monoclonal antibody of the IgG1 isotype that targets netrin-1 (NTN1), a protein overexpressed in many human cancers. By binding to and neutralizing netrin-1, NP137 prevents its interaction with dependence receptors such as UNC5B receptor. This restores apoptosis in tumor cells and inhibits processes like epithelial-mesenchymal transition (EMT), thereby reducing tumor growth, metastasis formation, and resistance to chemotherapy or immunotherapy. Preclinical studies have shown anti-cancer effects both as monotherapy and in combination with other treatments. The drug is being developed primarily for solid tumors including pancreatic cancer (for which it has received FDA Orphan Drug Designation), endometrial cancer, cervical cancer, liver cancer, acute myeloid leukemia, and is also under preclinical investigation for endometriosis[1][2][3][4][5][6][7][8].
