Overview

NP159 is a monoclonal antibody that acts as a Siglec-15 inhibitor. It was developed by NextCure and serves as a surrogate murine antibody for NC605, which is being investigated for the treatment of osteogenesis imperfecta (OI), also known as brittle bone disease. OI is characterized by high bone turnover, abnormal bone formation, fragility, and recurrent fractures. Unlike current anti-resorptive treatments that inhibit both bone loss and formation (leading to increased density but poor quality), Siglec-15 inhibition with agents like NP159 aims to both inhibit bone loss and enhance osteoblast recruitment—resulting in new bone formation with improved mechanical properties. Preclinical studies in OI mouse models have shown that treatment with NP159 reduces fracture incidence and improves various measures of bone quality[1][6][7].
