Overview

NP234 is a natural compound-derived small molecule inhibitor of glycolysis being developed as an antiparasitic agent for the treatment of leishmaniasis. Identified through an integrated in-silico and in-vitro high-content screening pipeline of secondary metabolites, NP234 targets the glycolytic pathway of *Leishmania donovani* parasites. In preclinical studies, it demonstrated potent in-vitro activity against the intracellular amastigote stage of *L. donovani* with an $\text{IC}_{50}$ of 356 nM, showing high specificity and stability in molecular dynamics simulations without significant cytotoxicity in mammalian cell lines.
