Overview

NP2567 is a first-in-class, potent, and selective small molecule inhibitor of the DNA mismatch repair (MMR) protein PMS2, currently in preclinical development by NeoPhore. By targeting the MutLα complex (comprising MLH1 and PMS2), NP2567 aims to transiently inhibit the MMR pathway in tumor cells. This inhibition leads to the accumulation of DNA replication errors and the generation of novel neoantigens, thereby increasing the tumor mutational burden (TMB). The therapeutic strategy is designed to transform immunologically "cold" tumors into "hot" tumors, potentially sensitizing them to immune checkpoint inhibitors or other immunotherapies.
