NPB (nitrated form of phenylbutyrate) is a novel nitric oxide donor synthesized from chlorambucil. It is being developed as a potential chemotherapeutic agent for pancreatic cancer. NPB induces cell death in human pancreatic cancer cells primarily through necrosis, rather than apoptosis, and also causes S-phase cell cycle arrest. Its design leverages the high affinity of phenylbutyrate to human serum albumin (HSA) to achieve sustained blood retention and selective accumulation in tumor tissue via the enhanced permeation and retention (EPR) effect, allowing for gradual nitric oxide release.
