Overview

NPB40 is a monoclonal antibody (mAb) that targets B7-H3 (CD276), a transmembrane protein and member of the B7 family of immune checkpoint molecules. Originally identified through a screen of antibodies against human embryonic stem cells (hESCs), NPB40 was found to bind specifically to hepatocellular carcinoma (HCC) cells while showing minimal binding to primary hepatocytes. B7-H3 is recognized as a potential marker for cancer stem cells (CSCs) in HCC, and NPB40 binding is associated with increased expression of CSC markers and clonogenic survival. Mechanistically, NPB40 induces the internalization of B7-H3 and can mediate anti-tumor activity through antibody-dependent cellular cytotoxicity (ADCC), particularly in its chimeric form (Chi-NPB40). Preclinical studies have demonstrated that NPB40 can suppress tumor growth in HCC xenograft models, both as a standalone agent and when utilized for targeted delivery of cytotoxic payloads via antibody-drug conjugate (ADC) strategies.
