NPC-31 (also known as P092) is a small molecule drug candidate being developed by Nobelpharma for the treatment of prion diseases, such as Creutzfeldt-Jakob disease (CJD). The compound is a maleic acid salt of prochlorperazine, a phenothiazine derivative traditionally used as an antipsychotic and antiemetic. In the context of neurodegenerative prion diseases, NPC-31 is being repurposed for its potential to inhibit the conformational conversion of the normal cellular prion protein (PrPC) into the pathogenic, misfolded scrapie isoform (PrPSc). By preventing the accumulation of these toxic protein aggregates and potentially promoting their clearance, NPC-31 aims to slow the progression of these fatal neurodegenerative conditions. It is currently undergoing Phase 1/2 clinical trials in Japan.
