Overview

NPL-2008 is a brain-penetrant small molecule antagonist of Toll-like receptor 2 (TLR2) being developed by NeuroPore Therapies for the treatment of neurodegenerative diseases. TLR2 is a pattern recognition receptor that plays a central role in the innate immune response and neuroinflammation. In conditions like Parkinson's disease and Amyotrophic Lateral Sclerosis (ALS), TLR2 is upregulated on microglia and is activated by misfolded proteins such as alpha-synuclein, leading to chronic inflammation and neuronal damage. By blocking TLR2 signaling, NPL-2008 aims to suppress neuroinflammatory pathways, reduce microglial activation, and potentially slow disease progression. The compound is currently being evaluated in early-stage clinical trials to assess its safety, tolerability, and pharmacokinetics.
