NPM1-A2 TCR-T cells are an autologous T-cell receptor (TCR) engineered T-cell therapy designed to target acute myeloid leukemia (AML) cells harboring mutations in the nucleophosmin 1 (NPM1) gene. The therapy utilizes a TCR isolated from a healthy HLA-A*02:01-positive individual that specifically recognizes the NPM1-derived neoantigen CLAVEEVSL (and related variants like CMAVEEVSL and CQAVEEVSL) presented on HLA-A*02 molecules. By targeting these frameshift-derived neoantigens, which are highly specific to the leukemic clone and absent in healthy tissues, the therapy aims to induce potent and selective lysis of AML cells. It is currently being evaluated in a Phase I/II clinical trial for patients with relapsed or refractory NPM1-mutated AML.
