Overview

NPPB (5-nitro-2-(3-phenylpropylamino)benzoic acid) is a widely used, broad-spectrum chloride channel blocker and pharmacological research tool. It inhibits multiple chloride conductance pathways, including the cystic fibrosis transmembrane conductance regulator (CFTR), calcium-activated chloride channels (CaCCs / TMEM16), volume-regulated anion channels (VRACs), and ClC chloride channels (such as ClC-K2 and ClC-3). In addition to its chloride channel blocking activity, NPPB acts as an agonist of the G-protein coupled receptor GPR35 and exhibits protonophoric activity, uncoupling mitochondrial oxidative phosphorylation at higher concentrations. It is primarily utilized as a laboratory reagent for in vitro and ex vivo electrophysiological and ion transport studies.
