Drug intelligence / Profile preview
NPRC blocking peptide (also known as AZ12107657 or NPR3i) is a selective peptide inhibitor of the natriuretic peptide clearance receptor (NPRC, also known as NPR3). Developed by AstraZeneca in collaboration with Karolinska Institutet, the peptide blocks the clearance activity of NPRC, which is highly expressed by glomerular podocytes. By preventing the degradation of natriuretic peptides (NPs) such as atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP), and C-type natriuretic peptide (CNP), the inhibitor enhances local NP-signaling and increases intracellular cyclic GMP (cGMP) levels. In preclinical models of kidney disease, including diabetic nephropathy and glomerulonephritis, treatment with the NPRC blocking peptide has demonstrated reno-protective effects, particularly when combined with angiotensin receptor blockers like losartan, by reducing albuminuria, glomerulosclerosis, and podocyte loss.
