Overview

NPRL2 is a gene encoding the nitrogen permease regulator 2-like protein (also known as tumor suppressor candidate 4 or TUSC4), which is a catalytic component of the GATOR1 complex. The GATOR1 complex functions as an inhibitor of the amino acid-sensing branch of the mTORC1 pathway by acting as a GTPase-activating protein for RagA/B within heterodimeric Rag complexes. This inhibition leads to downregulation of mTORC1 signaling in response to amino acid depletion. In cancer research and therapy development, gene therapy using NPRL2 has shown promising preclinical results in inducing anti-tumor immunity—particularly in KRAS/STK11 mutant and anti-PD1 resistant non-small cell lung cancer (NSCLC)—by promoting dendritic cell-mediated antigen presentation and cytotoxic immune cell activation[6][8][10]. Genprex is developing an intravenous formulation using lipid nanoparticles (DOTAP-NPRL2) for systemic delivery.
