NPS-2143 is a small molecule calcilytic drug that acts as a potent and selective antagonist of the extracellular calcium-sensing receptor (CaSR). By inhibiting CaSR, it stimulates the release of parathyroid hormone (PTH), which has been explored as a potential treatment for osteoporosis. NPS-2143 was the first calcilytic compound developed and remains widely used in research on CaSR biology and in the design of newer calcilytics. It has also been investigated for its effects on conditions such as pulmonary hypertension, melanoma, and other diseases involving abnormal calcium signaling or PTH regulation. The drug is primarily investigational and not approved for clinical use[1][2][3][4].
