NPT-2042 is an orally available small molecule drug developed by NeuroPro Therapeutics as a novel bumetanide analogue for the treatment of epilepsy, including intractable and reflex forms, as well as absence epilepsy. Unlike traditional antiseizure medications that broadly suppress neuronal excitability (often causing cognitive and neurological side effects), NPT-2042 is designed to reduce abnormal hypersynchrony—the synchronized firing of neurons that underlies seizure activity—by targeting glial cells rather than neurons. The drug acts primarily as a sodium-potassium-chloride symporter inhibitor, similar to bumetanide but with modifications intended to avoid diuretic effects and improve tolerability for neurological use[1][4][5].
