NPT088 is a recombinant fusion protein drug candidate developed for the treatment of neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. It is based on the General Amyloid Interaction Motif (GAIM) technology and consists of a human IgG1 Fc fused to GAIM, enabling it to target multiple misfolded protein aggregates that share a common amyloid fold. Mechanistically, NPT088 binds to and disrupts the formation of toxic aggregates of amyloid-beta (Aβ), tau, and alpha-synuclein proteins—key pathological features in Alzheimer's and Parkinson's diseases. Preclinical studies demonstrated that systemic administration of NPT088 reduced Aβ plaque, tau pathology, alpha-synuclein aggregation, brain atrophy, and improved cognitive performance in animal models. In early clinical trials (Phase 1), it was generally safe with infusion reactions as the most notable adverse event; however, no significant effect on brain plaques or symptoms was observed in patients with mild-to-moderate Alzheimer's disease. Development has since been discontinued for both Alzheimer’s disease and Parkinson’s disease[1][2][3][4][5][6][7].
