Overview

**NPT1220-312** is a potent, selective, orally bioavailable small molecule antagonist targeting Toll-like receptor 2 (TLR2) and Toll-like receptor 9 (TLR9). Developed by Neuropore Therapies, it acts as an allosteric non-competitive inhibitor, potently blocking TLR2- and TLR9-mediated pro-inflammatory signaling, cytokine release (e.g., IL-6, IL-1β, IL-18), chemokine production, and NLRP3 inflammasome activation in human microglia, macrophages, and whole blood. It demonstrates efficacy against both synthetic and natural TLR2 agonists, with IC50 values in the low nanomolar range (e.g., 260 nM for TLR1/2, 470 nM for TLR2/6, 317 nM for TLR9), and is designed for neurodegenerative disorders driven by neuroinflammation.[1][2][4]
