Overview

NPT1220-478 is a potent and selective small molecule dual antagonist of Toll-like receptor 2 (TLR2) and Toll-like receptor 9 (TLR9). It is being developed as a disease-modifying therapeutic for neurodegenerative disorders, particularly Parkinson’s disease. By blocking TLR2 and TLR9, NPT1220-478 aims to reduce neuroinflammation and the accumulation of neurotoxic protein aggregates, processes implicated in the progression of diseases such as Parkinson’s. Preclinical studies have demonstrated that NPT1220-478 reduces markers of inflammation and neurotoxic protein burden in multiple cell systems, including patient-derived neurons from individuals with Parkinson's disease. The drug has shown promising safety and pharmacokinetic profiles in preclinical models, with plans for clinical development focused on both central nervous system (CNS) disorders like Parkinson’s disease and peripheral inflammatory conditions[1][2][3][5].
