NPT288 is a first-in-class recombinant fusion protein developed by Proclara Biosciences (formerly NeuroPhage Pharmaceuticals) for the treatment of neurodegenerative diseases associated with protein misfolding, such as Alzheimer's disease. The molecule consists of a General Amyloid Interaction Motif (GAIM), derived from the M13 bacteriophage, fused to a human IgG1 Fc fragment. NPT288 utilizes a "pan-amyloid" approach, where the GAIM domain specifically recognizes and binds to the common cross-beta sheet structure shared by various misfolded proteins. This binding triggers the disassembly of existing toxic aggregates and prevents the formation of new ones. While NPT288 successfully completed Phase 1 clinical testing, demonstrating safety and proof of target engagement, Proclara Biosciences subsequently shifted its focus to NPT189, a next-generation candidate with optimized properties.
