Overview

NPX267 is a first-in-class monoclonal antibody developed for the treatment of solid tumors that express HHLA2 (also known as B7-H7), a tumor antigen upregulated in many cancers independently of PD-L1. The drug targets and blocks killer cell immunoglobulin-like receptor 3DL3 (KIR3DL3), an inhibitory receptor found on exhausted T cells and natural killer (NK) cells. By inhibiting the interaction between KIR3DL3 and HHLA2, NPX267 aims to reverse immune suppression within the tumor microenvironment, reactivate tumor antigen-primed immune cells, and enhance anti-tumor immunity. This approach is particularly promising for patients whose tumors are PD-L1-negative but express high levels of HHLA2[1][4][5][8].
