NPX887 is a fully human, antagonistic immunoglobulin G1 (IgG1) monoclonal antibody developed by NextPoint Therapeutics for the treatment of solid tumors. It targets HHLA2 (also known as B7-H7), a novel immune checkpoint and tumor antigen highly expressed in many cancers independently of PD-L1. By binding to HHLA2, NPX887 blocks its interaction with killer cell immunoglobulin-like receptor 3DL3 (KIR3DL3), thereby inhibiting KIR3DL3-mediated immunosuppression and preventing immune escape in solid tumors. This mechanism is believed to promote T cell and natural killer (NK) cell antitumor activity within the tumor microenvironment. The drug is currently being evaluated in Phase 1 clinical trials for multiple solid tumor types, including non-small cell lung carcinoma (NSCLC), small cell lung carcinoma (SCLC), renal cell carcinoma (RCC), colorectal carcinoma (CRC), and other malignancies expressing HHLA2/B7-H7[1][2][5][6][8].
