NQO1-AS gapmer is an antisense oligonucleotide (specifically a chimeric gapmer) designed to target and knock down NQO1-AS (NQO1 antisense RNA), a long non-coding RNA (lncRNA) transcribed from the antisense strand of the 3' UTR of the NQO1 gene. In metastatic breast cancer, NQO1-AS is upregulated and forms a stable duplex with NQO1 mRNA, stabilizing it and increasing the expression of the cytoprotective redox enzyme NADPH quinone dehydrogenase 1 (NQO1). This metabolic reprogramming protects cancer cells from oxidative stress and ferroptosis, facilitating metastatic colonization (particularly in the lungs). By utilizing NQO1-AS gapmers to knock down NQO1-AS, researchers can destabilize NQO1 mRNA, reduce NQO1 protein levels, and sensitize metastatic breast cancer cells to oxidative damage and ferroptosis, thereby inhibiting metastatic progression.
