NR2F6 siRNA (tCellVax) is an investigational RNA interference (RNAi) therapy developed by Regen BioPharma to target the NR2F6 (Nuclear Receptor Subfamily 2 Group F Member 6) nuclear receptor. NR2F6 acts as an intracellular immune checkpoint in T cells, and its expression is associated with immune evasion and tumor progression. By utilizing a proprietary siRNA molecule to silence NR2F6, the drug aims to induce T-cell hyper-responsiveness and enhance anti-tumor immune activity, characterized by the increased secretion of cytokines such as IFNγ, IL-2, IL-17, and IL-18. Regen BioPharma has explored NR2F6 siRNA as a monotherapy and in combination with PD-1/PD-L1 inhibitors for various malignancies, including endometrial cancer, and has submitted an Investigational New Drug (IND) application to the FDA for clinical evaluation. Additionally, research presented at AACR 2025 highlights the potential of NR2F6 siRNA to disrupt the NR2F6-NACC1-NOTCH3 axis in endometrial cancer, reducing tumor proliferation and therapy resistance.
