Nr4aTKO CD19-targeted CAR T cells are an experimental cellular immunotherapy designed to overcome the limitations of CAR T-cell therapy in solid tumors, specifically the state of T-cell exhaustion or hyporesponsiveness. These cells are engineered to express a chimeric antigen receptor (CAR) specific for the CD19 antigen and are further modified by the triple knockout (TKO) of the Nr4a transcription factor family: Nr4a1 (Nur77), Nr4a2 (Nurr1), and Nr4a3 (Nor1). Research conducted at the La Jolla Institute for Immunology has identified these Nr4a factors as key mediators of the exhaustion program downstream of NFAT signaling. By deleting these factors, the CAR T cells exhibit enhanced effector functions, including increased production of granzymes and cytokines, and demonstrate superior tumor regression and survival in preclinical models of solid tumors expressing CD19.
